    Overview

    GHRP-6 (Growth Hormone Releasing Peptide-6) is a synthetic hexapeptide with the sequence His-D-Trp-Ala-Trp-D-Phe-Lys-NH2. It is an agonist of the ghrelin receptor (GHS-R1a), also known as the growth hormone secretagogue receptor, and stimulates growth hormone secretion from the pituitary gland in research models.

    Synergy with GHRH

    A noted characteristic of GHRP-6 in laboratory studies is its synergistic interaction with GHRH. When studied together, the combined action of these peptides on GH release has been observed to be greater than the sum of their individual effects, a phenomenon attributed to their distinct receptor mechanisms.

    Mechanism of Action

    GHRP-6 binds to and activates the growth hormone secretagogue receptor type 1a (GHS-R1a), the same receptor activated by the endogenous hormone ghrelin. This receptor activation initiates intracellular signaling cascades that have been shown to stimulate GH synthesis and secretion from somatotroph cells in the anterior pituitary.

    Secondary Hormonal Effects

    The peptide has also been observed to have some effect on cortisol and prolactin secretion, which is attributed to the presence of ghrelin receptors on corticotroph and lactotroph cells.
